Want to confirmation with HR about the resignation process because i'm fresh grad

I want to ask, is it the HR will check with the employee's family member if the employee want to resign, such example the employee told the HR that they want to resign because of they got another job or they told the HR that their mom is sick and need to allocate to their own hometown, but in fact their mother's is passed away. Is it HR will validate first before terminate with the employee or since the Manager has acknowledge the resign letter, HR no need to validate first
